چکیده
Vascular dementia (VD) is a clinical syndrome of acquired cognitive dysfunction caused by various cerebrovascular factors. Estrogen steroid hormone involved in promoting neuronal survival and regulating many signaling pathways. However, the mechanism which it confers neuroprotective effects VD remains unclear. Here, we aimed to investigate effect estrogen on injury impairment rats. Adult female rats were randomly divided into four groups (sham, model, early later treatment) received sham surgery or bilateral ovariectomy permanent occlusion common carotid arteries (BCCAO). The treatment group daily intraperitoneal injections 17?-estradiol (100 µg/kg/day) for 8 weeks starting day after BCCAO. was administered same 1 week Learning memory functions assessed using Morris water maze. Morphological changes within hippocampal CA1 region observed hematoxylin/eosin staining electron microscopy. Expression proteins associated with autophagy detected immunohistochemical Western blot. We found that significantly alleviated damage reduced expression Beclin1 LC3B, indicating suppression autophagy. Moreover, enhanced ?-catenin Cyclin D1, while reducing glycogen synthase kinase 3?, suggesting activation Wnt/?-catenin signaling. These results indicate ameliorates learning deficiencies rats, this may be explained

The Canonical Wnt Signaling (Wnt/β-Catenin Pathway): A Potential Target for Cancer Prevention and Therapy
Precise regulation of signal transduction pathways is crucial for normal animal development and for maintaining cellular and tissue homeostasis in adults. The Wnt/Frizzled-mediated signaling includes canonical and non-canonical signal transduction pathways. Upregulation or downregulation of the canonical Wnt-signaling (or the Wnt/β-Catenin signal transduction) leads to a variety of human diseas...
متن کاملAngiopoietin-like protein 8 (betatrophin) may inhibit hepatocellular carcinoma through suppressing of the Wnt signaling pathway
Objective(s): Hepatocellular carcinoma (HCC) is one of the leading fatal neoplasms and the most common primary liver malignancy worldwide. Peptide hormone ANGPTL8 (betatrophin) may act as an important regulator in HCC development through the Wnt/β-catenin pathway. We aimed to evaluate the effects of recombinant ANGPTL8 on Wnt/β-catenin signaling in human liver carcinom...
متن کاملBHX Inhibits the Wnt Signaling Pathway by Suppressing β-catenin Transcription in the Nucleus
BHX (N-(4-hydroxybenzyl)-1,3,4-triphenyl-4,5-dihydro-1H-pyrazole-5-carboxamide), a Wnt signaling pathway inhibitor, effectively inhibits tumor cell growth, but the underlying mechanism is unclear. Thus, we aim to investigate the effects and associated mechanism of BHX action on A549 and MCF-7 cell lines. In our study, MTT(3-[4,5-dimethyl-2-thiazolyl]-2,5-diphenyl-2H-tetrazolium bromide) and xen...
متن کاملCelecoxib and 2,5‐dimethylcelecoxib inhibit intestinal cancer growth by suppressing the Wnt/β‐catenin signaling pathway
We previously reported that celecoxib, a selective COX-2 inhibitor, strongly inhibited human colon cancer cell proliferation by suppressing the Wnt/β-catenin signaling pathway. 2,5-Dimethylcelecoxib (DM-celecoxib), a celecoxib analog that does not inhibit COX-2, has also been reported to have an antitumor effect. In the present study, we elucidated whether DM-celecoxib inhibits intestinal cance...
متن کاملThe Role of Wnt/β-catenin Signaling Pathway in Rat Primordial Germ Cells Reprogramming and Induction into Pluripotent State
Primordial Germ Cells (PGCs) are unipotent precursors of the gametes. PGCs can give rise to a type of pluripotent stem cells in vitro that are called embryonic germ (EG) cells. PGCs can also acquire such pluripotency in vivo and generate teratomas. Under specific culture conditions, PGCs can be reprogrammed to embryonic germ cells which are capable of expression of key pluripotency marker...
